Standard Operating Procedure: Quantitative HPLC Purity Determination of Synthetic Polypeptides
Chromatographic purity determination requires strict adherence to system suitability parameters as outlined in European Pharmacopoeia (Ph. Eur.) Chapter 2.2.29 and USP <621>. Stationary phase selection typically utilizes octadecylsilyl (C18) chemically bonded porous silica microparticles (particle diameter: 1.7 to 3.0 µm, pore size: 100 to 300 Å) configured in 2.1 × 150 mm columns.
The mobile phase architecture comprises a binary aqueous-organic gradient system. Solvent A consists of 0.1% (v/v) ultra-pure trifluoroacetic acid (TFA) in Milli-Q grade water (resistivity > 18.2 MΩ·cm at 25°C). Solvent B consists of 0.1% (v/v) TFA in LC-MS grade acetonitrile. Flow rate is maintained at 0.300 mL/min with thermostated column temperature at 40.0 ± 0.2°C.
Sample concentration: 1.0 mg/mL in 10% Aqueous Acetonitrile
Injection volume: 5.0 µL (Autosampler tray maintained at 4.0°C)
UV Detector: Diode Array Detector (DAD) monitored at 214.4 nm (bandwidth 4 nm, ref. off)
Gradient profile: 5% B (0-2 min) -> 65% B (2-22 min) -> 95% B (22-25 min) -> 5% B (25-30 min)
Theoretical plate count (N): > 12,000 | Peak symmetry factor (As): 0.95 - 1.25
All calculated purity values represent the integrated peak area percentage of the principal compound relative to the total area of all detectable peaks, excluding mobile phase background artifacts and solvent blank signals.
